Description
SR9011 is a synthetic REV-ERBα/β agonist, structurally related to SR-9009 (Stenabolic), developed to explore the role of circadian regulation, mitochondrial activity, and lipid metabolism in health and disease. By binding to nuclear receptors REV-ERBα and REV-ERBβ, SR9011 modulates the expression of genes involved in metabolic homeostasis, inflammation, and energy utilization.
SR9011 is not a SARM and does not interact with androgen receptors. Instead, it influences metabolic and physiological processes at the genomic level, particularly through pathways linked to the biological clock, fat oxidation, and muscle endurance.
In preclinical studies, SR9011 has been observed to:
- Enhance mitochondrial biogenesis and oxidative metabolism in skeletal muscle tissue
- Increase total energy expenditure and attenuate adipose accumulation under sedentary conditions
- Improve exercise tolerance and endurance-related parameters in animal models, independent of physical training
- Suppress hepatic lipogenesis and reduce plasma lipid levels, including cholesterol and triglycerides
- Attenuate pro-inflammatory cytokine expression and modulate innate immune signaling pathways, with implications for autoimmune and metabolic disease research
- Alter the expression of circadian clock genes, affecting temporal regulation of metabolic and behavioral rhythms
Compared to SR-9009, SR9011 often exhibits greater potency and selectivity in cell-based models. However, like SR-9009, it suffers from limited bioavailability, which restricts systemic in vivo application without specialized delivery systems.
