SR-9009 (Stenabolic) is a synthetic agonist of the REV-ERBα/β nuclear receptors. Although not classified as a selective androgen receptor modulator (SARM), it is frequently included in research investigating metabolic and endurance-related pathways due to its significant effects on cellular energy regulation. SR-9009 engages REV-ERB receptors, which are integral to the regulation of circadian rhythms, mitochondrial biogenesis, lipid metabolism, and inflammatory responses.

Activation of REV-ERBα by SR-9009 modulates the transcriptional activity of genes implicated in energy expenditure, glucose and lipid homeostasis, and mitochondrial oxidative capacity. These mechanisms have made SR-9009 a molecule of interest in preclinical models of metabolic disorders, including obesity, dyslipidemia, and reduced exercise capacity.

In preclinical studies using murine models, SR-9009 has been shown to:

• Enhance endurance and exercise capacity via upregulation of mitochondrial biogenesis and oxidative metabolism in skeletal muscle
• Promote glucose uptake and reduce adiposity independent of caloric restriction or physical activity
• Suppress de novo lipogenesis and cholesterol biosynthesis, leading to improved lipid profiles

• Influence circadian gene expression, suggesting potential applications in models of sleep disturbance or metabolic dysregulation

Notably, SR-9009 does not interact with androgen receptors and lacks the hormonal activity associated with androgen modulators. It does not induce testosterone suppression, nor does it necessitate post-administration hormonal intervention. However, its limited oral bioavailability and rapid systemic clearance have led to continued research into structurally related analogs and optimized delivery modalities to enhance its pharmacokinetic profile.
